Influence of glucose antimetabolites on the Walker tumor.

چکیده
A large number of antimetabolites of nucleic acid components have been tried as antitumor agents (11). While some have retarded tumor growth, they are in general toxic to the host ani mal. This is perhaps to be expected, since protein synthesis is an essential phenomenon in all tissues. In the studies to be described, we have at tempted to interfere with the energy sources of the cell. Since the utilization of glucose by the glycolytic pathway appears to be more important for the malignant than the normal cell, the effects of glucose antimetabolites on tumor growth were studied at dosage levels which were not obviously toxic to the animal. Glucosamine has been tried as an antitumor agent by a number of workers, with inconclusive or negative results (2, 4, 6-8, 12, 13, 15-17) after the original promising observations of Quastel and Cantero (10). In most cases, however, a single daily dose of the test material was used. 2-DeoxyD-glucose (2DG) appears to have produced some suppression of tumor growth or glycolysis (5, 14, 20, 21). Since compounds of this type are readily excreted by the kidney, effective levels of the antimetabolite were probably present for only a very short time. In the present studies, we have used multiple daily doses of glucosamine and 2-deoxyD-glucose to achieve a more constant exposure to the compounds. The amount of glucosamine em ployed was generally at least double that reported by other observers. Both compounds produced sig nificant reductions in tumor growth at levels which did not seriously affect the health of the host.

Glucose Influence on Copper Ion-Dependent Oxidation of Low Density Lipoprotein
Background: It is well established that oxidative modification of low density lipoprotein (LDL) plays a causal role in human atherogenesis and the risk of atherosclerosis is increased in patients with diabetes mellitus. We examined the in vitro effect of glucose on native and glycated LDL oxidation using copper ion dependent oxidation system. Methods: In this study, LDL was isolated from plasma...
متن کاملAntimetabolite TTL-315 selectively kills glucose-deprived cancer cells and enhances responses to cytotoxic chemotherapy in preclinical models of cancer
Maintaining thiol homeostasis is an imperative for cancer cell survival in the nutrient-deprived microenvironment of solid tumors. Despite this metabolic vulnerability, a selective approach has yet to be developed to disrupt thiol homeostasis in solid tumors for therapeutic purposes. In this study, we report the identification of 2-mercaptopropionyl glycine disulfide (TTL-315) as a novel antime...
متن کاملEffects of Influence Parameters on Color Formation in Glucose Syrups during Storage
Effects of pH, temperature, and syrup concentration on color formation in glucose syrups were studied and the shelf life of syrups under various conditions was estimated. Temperatures of 5, 25, and 45 ºC, pH values of 4, 5, and 6 as well as concentrations of 30, 40, 50, 60, 70, and 80 ºBrix were examined. After 26 weeks no significant color changes were observed at 5 ºC. At 45 ºC, color formati...
متن کاملInfluence of salinity acclimatization on energy, oxygen consumption rate and glucose levels for Carassius auratus (Goldfish)
This study aimed to investigate the influence of increasing salinity concentrations on some indicators of energy consumption such as dissolved oxygen consumption, the consumed energy, as well as the levels of blood glucose in Carassius auratus (Goldfish), because of their direct relation with the metabolism rates when fish are exposed to exertion. Fish with an average weight of 37.5+2.5 g were ...
متن کاملEffects of 6-aminonicotinamide and 2-deoxy-D-glucose combination on the bioenergetics of perfused Ehrlich ascites tumour cells as monitored by 31 P-MR spectroscopy
The niacin antagonist, 6-aminonicotinamide (6-AN) is currently being evaluated in preclinical trials as a potential radiosensitizer. 2-Deoxy-o-glucose, a glucose antimetabolite is another promising differenti al radiomodifier. In this paper we report the effects of these antimetabolites on changes in bioenergetics of perfused immobilised Ehrlich ascites tumour (EAT) cells to predict their usefu...
